Donna Macmillan, Senior Scientist at Lhasa, presents on Lhasa’s work toward developing a Defined Approach to Skin Sensitisation Prediction.

A defined approach decision tree has been designed using exclusion criteria based on applicability domain knowledge, and limitations of Derek Nexus and in chemico/in vitro information sources covering key events 1-3 in the skin sensitisation Adverse Outcome Pathway (AOP). The hypothesis is that using exclusion criteria to de-prioritise less applicable assays and/or in silico outcomes produces a rational, transparent, and reliable defined approach for the prediction of skin sensitisation potential. After assessing a chemical against the exclusion criteria, guidance is provided to the most suitable in silico and in chemico/in vitro information sources. Results from the prioritised information sources are used in a ‘2 out of 3’ approach to provide a reliable hazard and potency prediction.
