Vitic 2022.1 data release: A high quality source for toxicity data

Over the past 12 months, a focus for Scientists at Lhasa Limited has been on developing genotoxicity and carcinogenicity endpoints in Vitic. Vitic aims to provide a wealth of high-quality data to support chemical risk assessments. Within this article, we explore some of the key improvements in the 2022.1 Vitic data release.

Vitic is a chemical database and information management system offering access to high-quality, expertly curated toxicity data from both published and proprietary sources.

Used as a solution by many of the world’s leading pharmaceutical, cosmetic, and chemistry-related organisations, Vitic offers rapid access to over 547,000 data records and is a quick and inexpensive way to identify potentially toxic chemicals and, perform read across during regulatory assessment.

We are pleased to announce the release of the Vitic 2022.1 database in February 2022. The 2022.1 database contains the following data:

  • New data records = 20,650
  • Total data records = 547,073
  • New substances = 1,006
  • Total substances = 22,849

A redesign of the Carcinogenicity table has improved access to, and display of, the carcinogenicity data, including improvements to the tumour-specific dose-response data. The addition of new studies has resulted in a total of 18,826 carcinogenicity records for 3,899 substances, with data being extracted from sources such as the NTP study reports and FDA novel drug approval documents. The addition of more study data has included the expansion of the nitrosamine dataset, which now includes 1,034 carcinogenicity study records for 262 nitrosamines.

Significant data extraction has been conducted for the Genetic Toxicity In-Vitro table. This table now contains a total of 304,231 records for 17,126 substances. New data was extracted from sources such as member-donated data, Joint Meeting on Pesticide Residues documents, Literature papers and FDA novel drug approval documents. The additional data has expanded the nitrosamine dataset, which now includes 7,080 in vitro genetox study records for 500 nitrosamines.

Data extraction also focussed on expansion of the Genetic Toxicity In-Vivo table dataset. This now contains 18,967 study records for 3,937 substances, with data extracted from sources such as Join Meeting on Pesticide Residues documents and FDA Novel Drug Approval documents. This has, in turn, expanded the nitrosamine dataset, which now includes 395 in vivo genetox study records for 42 nitrosamines.

In addition to data collection, work has also been done to condense the wealth of toxicity information available in Vitic into a more accessible format. The Genetox In-Vitro Summary table has been expanded to also include single summary calls for substances with Comet assay, HPRT assay, and Unscheduled DNA synthesis assay data (in addition to the Ames test, chromosome aberration test, micronucleus assay and mouse lymphoma assay already covered). As with previous assays, the data was curated to include the highest quality studies, then separated into subsets of similar assay conditions (e.g. cell line, metabolic activation and protocol). Each subset was assigned an activity call (positive, conflicted, equivocal or negative), with the subsets then being collated to give a summary call per substance. There are now 23,933 activity calls across 7 different assays for 16,644 substances.

In addition, a new table, the Genetox In-Vivo Summary table, was created. As with the Genetox In-Vitro Summary table, this was intended to enable more effective access to the Vitic data by providing a single summary call for each substance regarding a particular assay. Summary calls have now been generated for substances with in vivo chromosome aberration assay, comet assay, micronucleus assay, transgenic rodent assay, and unscheduled DNA assay data. Subsets were formed based on species, route of administration and organ, before being combined to a single summary call. A total of 3,858 summary calls have been generated for the 5 assays relating to 2,787 substances.
