Adverse outcome pathways for the nephrotoxicity of non-steroidal anti-inflammatory drugs.pdf
Non-steroidal anti-inflammatory drugs (NSAIDs), a class of compounds categorized as one of the most toxic in the FDA CDER urinary tract toxicity dataset, were investigated to identify the mechanisms leading to renal toxicity. Six structural toxicity alerts describing the nephrotoxicity of NSAIDs were implemented in the Derek Nexus knowledge base. Tentative AOPs to describe the in vivo mechanisms of renal toxicity for the generalized NSAID class were developed which show a dependence on the inhibition of the cyclooxygenase (COX) enzyme in renal tissues.
